Right well the plot is so bloomin' complicated that this'll take some summarising!. Here we go:

Leonardo Vetra is discovered by Maxamillion Kohler, murdered with the Illiminati symbol branded onto his chest and his eye torn out!. Kohler contacts Langdon and asks for his help finding the killer!.
Langdon deems the symbol authentic and the secret society active!.
It is revealed that the Illuminati has also stolen a small amount of deadly antimatter!. The canister’s electrical charge kept the antimatter safe but when stolen it runs on battery power which only lasts 24 hours and when it runs out of power the antimatter will explode with nuclear force!. The Illuminati have put the canister of antimatter somewhere in the Vatican with a camera pointing to its clock display, counting down!.
Langdon and Vittoria Vetra go to Vatican City where a new Pope is going to be elected soon!. Cardinal Mortati is in charge of the election and discovers that the four most senior cardinals most likely to become Pope are missing!. After they arrive, Langdon and Vittoria begin searching for them hoping that they will also find the antimatter!. They are helped by the late Pope’s aide, Camerlengo Carlo Ventresca and the Swiss Guard including Commander Olivetti, Captain Rocher and Lieutenant Chartrand!.
Langdon suspects the Illuminati are responsible for the cardinals’ disappearance and thinking of the means of induction to the Illuminati, retraces the steps of the so-called "Path of Illumination” which prospective candidates for the Illuminati had to follow!. It consists of a series of small clues left in various landmarks in and around the Vatican City which led to the secret meeting place of the Illuminati!.
The Path leads Langdon to four major locations in Rome, each associated with the four primordial elements: Earth, Air, Fire, and Water!. When he gets to each location, Langdon finds one of the four Cardinals murdered in a way that relates to the element connected to the location e!.g buried for Earth, pierced lungs for Air, burned alive for fire and drowned for water!. After finding the bodies of the first two, Langdon hurries to the Santa Maria della Vittoria Basilica and finds the Cardinals’ murderer setting the third on fire!. The man also killed Leonardo Vetra and stole the antimatter!. He is a Hassassin under command of the anonymous Illuminati master known as “Janus"!. The Hassassin kills Commander Olivetti and almost kills Langdon and then kidnaps Vittoria!. Langdon escapes and gets to the final location but can’t save the fourth Cardinal!.
Following the Path of Illumination to rescue Vittoria, Langdon ends up at an abandoned castle-like structure with an underground tunnel leading straight to the Pope's chambers in the Vatican!. Langdon frees Vittoria, and together they kill the Hassassin!. They go back to St!. Peter's Basilica, where Kohler has arrived to confront the Camerlengo!. Langdon and Vittoria fear that Kohler is Janus, and that he has come to murder the Camerlengo as the final step in his plot against the Church!. Hearing the Camerlengo scream in agony, the Swiss Guards burst into the room and open fire on Kohler!. Just before he dies, Kohler gives Langdon a videotape that he claims will explain everything!.
The Swiss Guard begins to evacuate the Basilica!. As he is exiting the church, the Camerlengo apparently goes into a trance and rushes back into the Basilica, claiming that he has received a vision from God revealing the location of the antimatter!. The Camerlengo ventures into the Basilica catacombs and finds the canister tucked away in a small box!. Langdon and the Camerlengo retrieve the antimatter and get in a helicopter with only 5 minutes to spare!. The Camerlengo abandons Langdon in the helicopter and takes the only parachute to land safely onto the roof of St!. Peter's just as the canister explodes harmlessly in the sky!. The crowd in St!. Peter's Square don’t realise the Camerlengo used a parachute and think it’s a miracle that he is standing on the roof, so the Papal Conclave elect him as Pope!. Langdon survived the explosion by using a window cover from the chopper as a parachute and landed in the Tiber River near Tiber Island, which is famous for its reputation as an island blessed with miracles of healing!.
Langdon watches Kohler's tape with the College of Cardinals!. Langdon, Vittoria, and the cardinals confront the Camerlengo, revealing that before the events of the novel, the Pope was going to meet with Leonardo Vetra about his research at CERN!. Vetra believed that science was capable of establishing a link between Man and God, a belief that was manifested by his research on antimatter!. The Camerlengo didn’t like this and believed that the Church alone, not science, should dictate the moral beliefs of the Christian faith!. While discussing Vetra, the Pope reveals his support is due to science having created him a miracle: a son conceived by artificial insemination!. Horrified that the Pope has fathered a child, the Camerlengo plots to "rectify" the situation!. He poisoned the Pope and, under the guise of an Illuminati master (Janus), he recruited the Hassassin to kill Vetra and the four Cardinals and steal the antimatter!. The Camerlengo planted the antimatter in St!. Peter's and feigned his last-minute "vision" from God in order to be seen as a hero and the savior of Christendom by those who witnessed his brave acts!. The Illuminati’s involvement was therefore just a plot engineered by the Camerlengo to cover his own plans!. As Langdon suspected from the very beginning, the Order of the Illuminati has been dead for a long time!.
Finally, it’s revealed that Camerlengo Ventresca was the Pope’s artificially inseminated son, so overcome with grief and guilt especially from killing his own father, Camerlengo Ventresca soaks himself in oil and immolates himself before a crowd of onlookers in St!. Peter's Square!. The conclave elects Cardinal Mortati as the new pope!.
Langdon and Vittoria retire to the Hotel Bernini as lovers!. Lieutenant Chartrand delivers a letter and package to Langdon from the new Pope!. The package is the rare 'Illuminati Diamond' brand, which is loaned indefinitely to Langdon!.

Robert Langdon is the main protagonist and is a professor of Symbology at Harvard University!. He is convinced to fly to CERN to help investigate the murder of Leonard Vetra!.

Leonardo Vetra was a scientist working at CERN and a priest!. He was researching antimatter when he was murdered by the Hassassin!. He is also the adoptive father of Vittoria!.

Vittoria Vetra, the adopted daughter of Vetra, works with CERN!. She researches biology and physics!. She worked with her father in their research of antimatter!.

Camerlegno Carlo Ventresca is the Papal Chamberlain during the conclave and is eventually revealed to be the main antagonist in the novel!. He was responsible for the Pope’s murder!. He is also known as Janus!.

Cardinal Saverio Mortati is the most senior cardinal in the conclave!. He was the Devil's Advocate for the late pope and knows that the pope had a child!. He end up as the new Pope!.

Commander Olivetti was the commandant of the Swiss Guard!. Along with other Swiss Guards he searches for the missing antimatter hidden somewhere at the Vatican!. He is murdered by the Hassassin!.

Captain Rocher was the second in command after Commander Olivetti!. He is contacted by Max Kohler telling his knowledge on the real cause of the events!. He is killed by Lt!. Chartrand who thought he was with the Illuminati!.

The Hassassin was the killer hired by the Camerlegno!. He is of Middle Eastern origin and displays his sadistic lust for women throughout the novel!. He murders Leonardo Vetra, the Preferiti, and Commander Olivetti!. He is pushed from a balcony by Langdon and is killed by the fall!.

Maximilian Kohler was the wheelchair-bound director of CERN!. He contacts Langdon to help him find the killer of his friend, Leonardo Vetra!. He also blames religion as the reason for his disability and devotes his life to science!. He is killed by the Swiss Guard thinking he branded the Camerlegno, while in fact, Kohler was confronting the Camerlegno as his friend’s murderer!.

Universal themes could be mystery, religion, religion versus science, deception or murder!.
