Basically, as with all things, acting is a skill that is helped by experience, passion and a natural talent helps too. Remember that it isn't just the camera you are ignoring. to act on-screen you have to 'ignore' the camera, cameraman, sound people, director, costume and make-up people, other actors and sometimes random gawking 'audience' members. You must draw on personal experiences as well as your imagination to recreate the scene as if it were real.
Many people have written about different techniques and methods of acting, you can look for books on the subject in your library or try these sites:

So You Wanna (soyouwanna.com) also has step-by-step guidelines on being "discovered"

An agent can be very helpful in finding auditions. Look up talent agency in your Yellow Pages or look online for local agents.

One of the biggest grips is speaking. If you don't reharsh your lines enough to make them not sound like your rehashing a line you read thats exactly how it will sound. It needs to sound like a natural conversation in the tone of the environment. Now what is extremely hard is acting with a bluescreen or with CGI stand ins where what will be on film is not there when your preforming. Also some older filming tricks where actor size is changed and couldn't used split screen and before CGI. They would set up a perspective view shot where the larger character is closer to the camera then the smaller character, the two would have to interact as if in front of each other. Mat cell painting is another point where you would be reforing to something not there. These would be still paintings on glass set up in front of the camera to add layers of backdrop to a scene they couldn't build feasibly.
