It depends - if it's not by appointment, then yes.

If it's an amateur production, almost always.

If it's a college play, different colleges have different policies--and they'll always prefer their own students even if non-students are welcome to audition.

If it's a school play, no.

If it's a professional show, almost all of them are open only to members of one of the three actors' unions (Equity, SAG, AFTRA). A few professional shows do hold open auditions, and you may imagine the competition is brutal. There are also rarely some opportunities to show up for an open call for extras (say, a location shoot somewhere away from the usual media centers) where, if accepted, you'll get no lines, stand around most of the day, and then mingle or run or sit or whatever they want the crowd to do.
