I know that it sounds rather stupid, but READ the character. READ the play. READ the scene, if there is no real play. Make up the character. Write a character biography, including all of the random things that the character likes and doesn't like. Make the character real to YOU. Do this with different characters, use your imagination a little. It will help you, I promise. Once you "get to know them" it will be much easier.

I also might suggest starting off with someone you know. Write down what you know about them, what they like, etc. Then, try to act like them. It will help you, and even if it isn't completely real, you're still getting practice.

This is not a simple question that can be answered in a forum, for starters read a Challenge for the Actor by Uta Hagen that is my favorite book of all that i have read. She talks a lot about observation, you need to observe yourself and others, take what you learn from the observations and put it up on stage. Study your favorite actors, what do they do when they walk in a room, drink a cup of coffee, etc.?
