Know the script. Know the characters.
Have a vision of what you want to do with the script.
Be extremely organized.

I didn't read through this website, but I just googled Tips on directing a play. There were several websites.

Ask someone to be your stage manager who will hold the book-- the script copied out and stuck into a binder with each page enlarged to A4 and a blank page stuck in oppo so that notes can be made -- during rehearsals and jot down your movement, stage directions, cueing both FX and LX and make notes on bits of business you've discussed with actor. This frees you up to actually watch your actors and see how they're delivering lines, interacting with each other and moving. SM very helpful. Oh, and go through the play scene by scene and decide where your characters are needed, and when you've cross-referenced this info, then you can draw up a rehearsal schedule over the period you need. Sometimes only a couple of actors are wanted for a rehearsal, other times you want the entire ensemble. Break a leg.

Your question is too vague.... fill in some blanks please.... like what type of play is it? How long do you have to prepare? What is your prior expreience in directing? What is your experience as a stage actor? Etc.... All this will determine my answer. I hope I'm wrong if I assume you have no acting experience and no directorial experience. If this be the case, and this play of your's is of any substantial size......... All I can say is you've bitten off a huge chunk my friend and you're going to need lots of help pulling this off.
