To audition for a Broadway show, you usually have to be a member of Actors Equity, the US Theater Actors Union. So writing a letter to a Broadway theater won't get you an audition to be in a show (but it might get you a chance to go backstage after a show and meet some of the people involved in the production.)

There are some exceptions to this: some Broadway shows (for example, Rent and Spring Awakening) hold "open auditions" to find cast members who may not be members of Actors Equity yet. In these cases, you want to write to the casting agency involved (not to the theater, who will have no say in casting) to ask about open auditions (and when you find out when they are, be prepared to do your absolute best, because Broadway casting agents can be even stricter and meaner than Simon Cowell.)

Broadway is not a specific building or address. Most simply put, it is an area that has a bunch of theaters in it.
