How do i ink and color a drawing on the computer?

i have a wacom tablet as well as adobe cs3 and corel painter. i am wondering it its easier to ink it with pen and then scan it or if it would look better inked on the comp. if i ink it in photoshop can you give me some tips like which brush to use and such. and when i color it other than making a new layer and setting it to "multiply" are there any tips on how to make that look best, especially with blending colors.
