Thats a big question.
Lighting, well make sure you have your Key light source, some additional fill lights might be needed (every scenario is different) some back lights or rim lights for your talent, some lights in dark sports in the background here and there can help add some depth. Then indoor or outdoor? Make sure you have the correct light source, or least fake it the best you can with some gels and diffusion added to your light source where needed. Are you shooting this on film, digital, hd, etc. Man there are soooo many tricks and methods to lighting a set its impossible to tell them all here. If you want proper lighting hire a good DP and Gaffer.

General set up, well think of your shots you need to tell the story. Angles, moving, locked off, on and on and on. Then plan ahead how to be doing one shot, while preparing what you can for the next, planing is soooo important in production.
