Everything..everything about them..
like you said..

how they talk..
-tone
-the pitch
-pause
-power
-use of vocabulary (or lack of)
-pace
etc things like that


body languege
-how the sit..stand...
-how they act towards other characters
-are they touchy type of people..or do they like there space?


also other things..like there status..are they a boss owner of a huge company..is they are they stand tall..walk around like they own the place..well they like do! :p and there not very friendly with the "lower-class"..
