Yes, the Writers Guild of America can register your script. It is an online process and you will receive a registration number instantly. It only costs about $20 -$25. The online registration process takes you step-by-step. Just make sure you fill-out everything completely. They will mail you a certificate of registration.
(Plus, there is a point system with selling and/or getting your script produced that goes towards your eligibility to join the WGA.)

They also allow a certain amount of updates, but I am not sure what it is. They do recommend that if you make significant changes, you file a new registration.

You should do this IN ADDITION to filing for a copyright. If nothing else, it is further proof that it is yours.

Mail a copy to yourself and DON"T OPEN IT. The postmark will prove you wrote it first.

copyrights returns are taking over a year, but the registration is effective the date they stamp it in, usually a week after you send it.

If it's a TV or movie script the Writer's Guild has a registration service but I don't know the current fees.

It's good for 10 years.
