All very flawed with tragic or mysterious pasts, also, you can't go wrong with killing one of the main characters to get the story started. I find you can create a very unique mood in a story by telling the back-story of a character who's already dead. (also ties in nicely with your themes)

If you read "The Five People You Meet in Heaven", it is exactly about this topic.... a humble, ordinary man who feels he wasted his life (except for marrying his wife who died too early)... and after he dies (at the beginning of the story), he goes through a learning process to discover the meaning his life had.
