Well, I think it would be a very bad idea to adjust your sound post unless you are an experienced violin luthier/repair person. It is a very dangerous thing to do, and if the sound post falls, you'll have to go get it reset by a luthier/repair person. Setting a sound post is a very precise art that requires masterful experience.

However, if you are experienced, of course you should never do it with the strings tightened on the bridge. If you do it while the bridge is still applying pressure to the wood, if the soundpost falls, the pressure of the bridge can crack the wood or even break it and make it fall in, completely destroying the front of the insturment, and you'll end up with a horrendous, expensive, ugly, pitiful mess, and you'll have to get horribly expensive repair bills or a new violin.

In short, my advice is to get a specialist to do this, or if you are experienced, do it yourself.
